Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001297.5(CNGB1):c.988G>A (p.Val330Ile), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with CNGB1-related conditions. This variant is present in population databases (rs775599315, gnomAD 0.006%). This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 330 of the CNGB1 protein (p.Val330Ile). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CNGB1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
