Several computational tools predict a significant impact on normal splicing: Two predict the variant weakens a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00031 in 1613260 control chromosomes, predominantly at a frequency of 0.0046 within the South Asian subpopulation in the gnomAD database, including 4 homozygotes. The observed variant frequency within South Asian control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for a pathogenic variant in ITGA2B causing ITGA2B-Related Disorders phenotype. To our knowledge, no occurrence of c.1752+4C>T in individuals affected with ITGA2B-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported.
