NM_002161.6(IARS1):c.1475G>A (p.Gly492Glu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1475G>A (p.G492E) alteration is located in exon 15 (coding exon 14) of the IARS gene. This alteration results from a G to A substitution at nucleotide position 1475, causing the glycine (G) at amino acid position 492 to be replaced by a glutamic acid (E). Based on data from the Genome Aggregation Database (gnomAD) database, the IARS c.1475G>A alteration was observed in 0.0028% (8/282476) of total alleles studied, with a frequency of 0.006% (8/128904) in the European (non-Finnish) subpopulation. This amino acid position is highly conserved in available vertebrate species. The in silico prediction for the p.G492E alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
