Experimental studies have shown that this premature translational stop signal affects DCTN1 function (PMID: 32023010). Algorithms developed to predict the effect of variants on protein structure and function are not available or were not evaluated for this variant. This premature translational stop signal has been observed in individual(s) with distal hereditary motor neuropathy (PMID: 32023010). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Leu210Alafs*90) in the DCTN1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in DCTN1 cause disease.
