Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007144.3(PCGF2):c.46C>T (p.Leu16Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 16 of the PCGF2 protein (p.Leu16Phe). This variant is present in population databases (rs757069949, gnomAD 0.06%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with PCGF2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1926370). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
