Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.1783A>G (p.Ser595Gly), citing Ambry Variant Classification Scheme 2023: The p.S595G variant (also known as c.1783A>G), located in coding exon 16 of the MLH1 gene, results from an A to G substitution at nucleotide position 1783. The serine at codon 595 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
