Uncertain significance for Leber congenital amaurosis 6; Cone-rod dystrophy 13 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020366.4(RPGRIP1):c.138GGA[1] (p.Glu47del), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This variant, c.141_143del, results in the deletion of 1 amino acid(s) of the RPGRIP1 protein (p.Glu47del), but otherwise preserves the integrity of the reading frame. This variant has not been reported in the literature in individuals affected with RPGRIP1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
