Likely pathogenic for Cystic fibrosis — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000492.4(CFTR):c.710A>C (p.Gln237Pro), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CFTR c.710A>C (p.Gln237Pro) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251392 control chromosomes. c.710A>C has been observed in a presumed compound heterozygous state in at least 1 individual affected with adolescent onset Cystic Fibrosis (example, Lemson_2025). A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.709C>G, p.Gln237Glu), supporting the critical relevance of codon 237 to CFTR protein function. At least 1 publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect resulted in approximately 14% of normal chloride channel conductance relative to wild type (e.g., Bihler_2024). The following publications have been ascertained in the context of this evaluation (PMID: 38388235, 40159410). ClinVar contains an entry for this variant (Variation ID: 1921232). Based on the evidence outlined above, the variant was classified as likely pathogenic.
