NM_001042631.3(SDHAF1):c.116A>C (p.His39Pro) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: SDHAF1 c.116A>C (p.His39Pro) results in a non-conservative amino acid change located in the Complex 1 LYR protein domain (IPR008011) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.8e-05 in 165698 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.116A>C in individuals affected with Mitochondrial Complex 2 Deficiency, Nuclear Type 2 and no experimental evidence demonstrating its impact on protein function have been reported. Variant was observed in a compound heterozygous individual affected with spasticity and paraparesis (internal testing). One submitter has cited clinical-significance assessments for this variant to ClinVar after 2014 and has classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
