This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 49367, where G is replaced by A; at the protein level this means replaces arginine at residue 16456 with histidine — a missense variant. Submitter rationale: Variant summary: TTN c.41663G>A (p.Arg13888His) results in a non-conservative amino acid change located in the A-band region of the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 5.3e-05 in 243908 control chromosomes (gnomAD). This frequency is not higher than expected for a pathogenic variant in TTN causing Dilated Cardiomyopathy (5.3e-05 vs 0.00039), allowing no conclusion about variant significance. c.41663G>A has been reported in the literature in a sudden unexpected death case (Scheiper_2018). This report does not provide unequivocal conclusions about association of the variant with Dilated Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three ClinVar submitters (evaluation after 2014) cite the variant as benign, likely benign and uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
