Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015426.5(POC1A):c.161C>T (p.Ala54Val), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Not Available"). This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 54 of the POC1A protein (p.Ala54Val). This variant is present in population databases (rs761891339, gnomAD 0.01%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with POC1A-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
