This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 95320, where A is replaced by G; at the protein level this means replaces lysine at residue 31774 with glutamic acid — a missense variant. Submitter rationale: Variant summary: TTN c.87616A>G (p.Lys29206Glu) results in a conservative amino acid change located in the A-band region, Fibronectin type-III domain of the encoded protein sequence. Two of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 5.6e-05 in 248670 control chromosomes (gnomAD), predominantly at a frequency of 0.00012 within the Non-Finnish European subpopulation in the gnomAD database. To our knowledge, no occurrence of c.87616A>G in individuals affected with Limb-Girdle Muscular Dystrophy, Type 2J and no experimental evidence demonstrating its impact on protein function have been reported. Three ClinVar submitters have assessed the variant since 2014: two classified the variant as uncertain significance and one as likely benign. Based on the evidence outlined above, the variant was classified as uncertain significance.
