Submitter rationale: This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 221 of the DONSON protein (p.Trp221Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with microcephaly, short stature, and limb abnormalities syndrome (PMID: 31320746). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
