NM_001082971.2(DDC):c.299G>C (p.Cys100Ser) was classified as Likely pathogenic for Deficiency of aromatic-L-amino-acid decarboxylase by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This missense change has been observed in individual(s) with aromatic L-amino acid decarboxylase deficiency (PMID: 30952622, 31975548, 32409695, 33734312). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs200339845, gnomAD 0.0009%). This sequence change replaces cysteine, which is neutral and slightly polar, with serine, which is neutral and polar, at codon 100 of the DDC protein (p.Cys100Ser).
