Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015072.5(TTLL5):c.3794C>T (p.Ala1265Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1265 of the TTLL5 protein (p.Ala1265Val). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TTLL5-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
