Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005006.7(NDUFS1):c.1814A>T (p.Gln605Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 605 of the NDUFS1 protein (p.Gln605Leu). This variant is present in population databases (rs758981403, gnomAD 0.01%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with NDUFS1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1916376). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt NDUFS1 protein function with a negative predictive value of 80%.
