Uncertain significance for Vici syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020964.3(EPG5):c.6166C>G (p.Arg2056Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 2056 of the EPG5 protein (p.Arg2056Gly). This variant is present in population databases (rs116076204, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with EPG5-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
