This variant lies in the BAG3 gene (transcript NM_004281.4) at coding-DNA position 1100, where C is replaced by G; at the protein level this means replaces proline at residue 367 with arginine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the BAG3 gene. The P367R variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The P367R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In silico analysis predicts this variant is probably damaging to the protein structure/function. However, to our knowledge no studies have been performed to determine the functional effect of the P367R variant. Furthermore, this substitution occurs at a position that is not conserved across species.
