Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.9577A>G (p.Arg3193Gly), citing Ambry Variant Classification Scheme 2023: The p.R3193G variant (also known as c.9577A>G), located in coding exon 39 of the AKAP9 gene, results from an A to G substitution at nucleotide position 9577. The arginine at codon 3193 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
