Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.5234T>C (p.Val1745Ala), citing Ambry Variant Classification Scheme 2023: The p.V1745A variant (also known as c.5234T>C), located in coding exon 21 of the AKAP9 gene, results from a T to C substitution at nucleotide position 5234. The valine at codon 1745 is replaced by alanine, an amino acid with similar properties. This alteration has been reported as a secondary cardiac variant in an exome cohort; however, clinical details are limited (Ng D et al. Circ Cardiovasc Genet, 2013 Aug;6:337-46). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
