Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003361.4(UMOD):c.842A>G (p.Glu281Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 281 of the UMOD protein (p.Glu281Gly). This variant is present in population databases (rs543775691, gnomAD 0.05%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with UMOD-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
