Uncertain significance for Immunodeficiency 104 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002838.5(PTPRC):c.1929G>C (p.Lys643Asn), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with PTPRC-related conditions. This variant is present in population databases (rs188500261, gnomAD 0.0009%). This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 643 of the PTPRC protein (p.Lys643Asn).
