Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014014.5(SNRNP200):c.1462C>G (p.Leu488Val), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs771467567, gnomAD 0.003%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with SNRNP200-related conditions. This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 488 of the SNRNP200 protein (p.Leu488Val).
