Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_139057.4(ADAMTS17):c.873+1G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ADAMTS17 gene (transcript NM_139057.4) at the canonical splice donor site of the intron immediately after coding-DNA position 873, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 5 of the ADAMTS17 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in a shortened protein product. This variant is present in population databases (no rsID available, gnomAD 0.003%). Disruption of this splice site has been observed in individual(s) with Weill-Marchesani-like syndrome (PMID: 24940034). ClinVar contains an entry for this variant (Variation ID: 1909558). Studies have shown that disruption of this splice site results in skipping of exon 5, but is expected to preserve the integrity of the reading-frame (PMID: 24940034). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
