Uncertain significance for Neonatal-onset encephalopathy with rigidity and seizures — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152743.4(BRAT1):c.2257C>T (p.Pro753Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 753 of the BRAT1 protein (p.Pro753Ser). This variant is present in population databases (rs773060720, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRAT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
