Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005477.3(HCN4):c.88G>A (p.Glu30Lys), citing Ambry Variant Classification Scheme 2023: The p.E30K variant (also known as c.88G>A), located in coding exon 1 of the HCN4 gene, results from a G to A substitution at nucleotide position 88. The glutamic acid at codon 30 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
