Submitter rationale: p.Pro3824Ala (CCC>GCC): c.11470 C>G in exon 43 of the ANK2 gene (NM_001148.4). Although rare, mutations in the ANK2 gene have been reported previously in association with LQTS (Mohler P et al., 2003). The P3824A variant has not been published as a mutation or as a benign polymorphism to our knowledge. The P3824A variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The P3824A variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties.
