Uncertain significance for Pulmonary fibrosis and/or bone marrow failure, Telomere-related, 3; Dyskeratosis congenita, autosomal recessive 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001283009.2(RTEL1):c.1451C>T (p.Pro484Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 484 of the RTEL1 protein (p.Pro484Leu). This variant is present in population databases (rs786205700, gnomAD 0.006%). This missense change has been observed in individual(s) with pulmonary fibrosis (PMID: 25848748). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 190473). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
