Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032603.5(LOXL3):c.2259C>G (p.Ile753Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 753 of the LOXL3 protein (p.Ile753Met). This variant is present in population databases (rs763423249, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with LOXL3-related conditions. ClinVar contains an entry for this variant (Variation ID: 1903038). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
