Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002335.4(LRP5):c.2900C>T (p.Pro967Leu), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 967 of the LRP5 protein (p.Pro967Leu). This variant is present in population databases (rs780789533, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with LRP5-related conditions.
