Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001145358.2(SIN3A):c.1967C>G (p.Thr656Ser), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with SIN3A-related conditions. This variant is present in population databases (rs780797915, gnomAD 0.007%). This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 656 of the SIN3A protein (p.Thr656Ser).
