This variant lies in the TYR gene (transcript NM_000372.5) at coding-DNA position 446, where A is replaced by C; at the protein level this means replaces tyrosine at residue 149 with serine — a missense variant. Submitter rationale: Variant summary: TYR c.446A>C (p.Tyr149Ser) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251434 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.446A>C has been reported in the literature in individuals affected with Oculocutaneous Albinism, however, they also carried a second homozygous pathogenic variant in the TYR gene : c.346C>T, p.Arg116* suggesting a benign role for c.44A>C (Ashaat_2024). These report(s) do not provide unequivocal conclusions about association of the variant with Oculocutaneous Albinism. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
