Likely pathogenic — the classification assigned by GeneDx to NM_001605.3(AARS1):c.242A>C (p.Lys81Thr), citing GeneDx Variant Classification (06012015): The K81T variant has been previously reported in this individual (Simons et al., 2015). Functional studies show K81T mildly impaired aminoacylation activity in vitro and significantly reduced growth in yeast (Simons et al., 2015). K81T was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. K81T is a semi-conservative amino acid substitution that alters a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function.
