Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.5058+2T>C, citing Ambry Variant Classification Scheme 2023: The c.5058+2T>C intronic variant results from a T to C substitution two nucleotides after coding exon 19 in the AKAP9 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
