Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152424.4(AMER1):c.2797G>A (p.Glu933Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 933 of the AMER1 protein (p.Glu933Lys). This variant is present in population databases (no rsID available, gnomAD 0.02%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with AMER1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1898217). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
