Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_006258.4(PRKG1):c.202T>A (p.Leu68Met), citing Ambry Variant Classification Scheme 2023: The p.L68M variant (also known as c.202T>A), located in coding exon 1 of the PRKG1 gene, results from a T to A substitution at nucleotide position 202. The leucine at codon 68 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
