Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001349338.3(FOXP1):c.1285A>G (p.Met429Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 429 of the FOXP1 protein (p.Met429Val). This variant has not been reported in the literature in individuals affected with FOXP1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant is present in population databases (no rsID available, gnomAD 0.0009%).
