NM_001369.3(DNAH5):c.2461A>G (p.Asn821Asp) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N821D variant (also known as c.2461A>G), located in coding exon 17 of the DNAH5 gene, results from an A to G substitution at nucleotide position 2461. The asparagine at codon 821 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
