NM_007294.3(BRCA1):c.4358_4484del was classified as pathogenic by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria: A deletion of exon 13 (c.4358_4484) in the BRCA1 gene is predicted to disrupt the translation reading frame of the BRCA1 mRNA and result in the premature termination of BRCA1 protein synthesis. In the published literature, a similar deletion of exon 13 in the BRCA1 gene has been reported in multiple individuals or families with hereditary breast and/or ovarian cancer (PMID: 32629901 (2020), 22544547 (2012), 22006311 (2011), 20232141 (2011), 16793929 (2006)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic.
