NC_000016.10:g.(?_23603458)_(23603669_?)del was classified as Pathogenic for Familial cancer of breast by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change is a gross deletion of the genomic region encompassing exon 13 of the PALB2 gene. This deletion extends to the edge of the assayed region and the 3' boundary of this event is not known. Truncating gross deletions in PALB2 are known to be pathogenic. A similar deletion has been reported in the literature in an individual with breast cancer (PMID: 25099575). Exon 13 encodes two WD40-like repeat motifs in the PALB2 (partner and localizer of BRCA2) gene. WD40 motifs are required for interaction with the BRCA2 protein (PMID: 16793542, 25099575). This exonic deletion is likely to truncate the PALB2 protein and abolish its DNA repair function. For these reasons, this sequence change has been classified as Pathogenic.
