This variant lies in the KMT2B gene (transcript NM_014727.3) at coding-DNA position 1090, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 364 with lysine — a missense variant. Submitter rationale: Variant summary: KMT2B c.1090G>A (p.Glu364Lys) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 5.6e-05 in 162154 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1090G>A in individuals affected with Dystonia 28, Childhood-Onset and no experimental evidence demonstrating its impact on protein function have been reported.
