Likely pathogenic for Bardet-Biedl syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000001.10:g.(243542166_243579003)_(243579132_243581269)del, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the deletion of exon 14 in the SDCCAG8 gene. A presumed nomenclature of c.(1616+1_1617-1)_(1744+1_1745-1)del has been designated for the purposes of this classification. Although exact breakpoints of this deletion are not known, it is expected to result in a frameshift in the SDCCAG8 gene, a known mechanism of disease. The variant was absent in 21694 control chromosomes (gnomAD, structural variants dataset). To our knowledge, no occurrence of c.(1616+1_1617-1)_(1744+1_1745-1)del in individuals affected with Bardet-Biedl Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted a clinical-significance assessment for this variant to ClinVar after 2014 and classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as likely pathogenic.
