Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.863C>T (p.Thr288Ile), citing Ambry Variant Classification Scheme 2023: The p.T288I variant (also known as c.863C>T), located in coding exon 10 of the MLH1 gene, results from a C to T substitution at nucleotide position 863. The threonine at codon 288 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
