This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 916, where A is replaced by G; at the protein level this means replaces isoleucine at residue 306 with valine — a missense variant. Submitter rationale: The p.I306V variant (also known as c.916A>G), located in coding exon 8 of the PTEN gene, results from an A to G substitution at nucleotide position 916. The isoleucine at codon 306 is replaced by valine, an amino acid with highly similar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955). This variant demonstrated possible wild-type like intracellular protein abundance on one multiplex functional assay (Matreyek KA et al. Nat Genet, 2018 Jun;50:874-882).
