NM_002878.4(RAD51D):c.185C>G (p.Ser62Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the RAD51D gene (transcript NM_002878.4) at coding-DNA position 185, where C is replaced by G; at the protein level this means replaces serine at residue 62 with tryptophan — a missense variant. Submitter rationale: The p.S62W variant (also known as c.185C>G), located in coding exon 3 of the RAD51D gene, results from a C to G substitution at nucleotide position 185. The serine at codon 62 is replaced by tryptophan, an amino acid with highly dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.01% (greater than 11000 alleles tested) in our clinical cohort. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.S62W remains unclear.
