Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.3685G>A (p.Val1229Ile), citing Ambry Variant Classification Scheme 2023: The p.V1229I variant (also known as c.3685G>A), located in coding exon 10 of the BRCA2 gene, results from a G to A substitution at nucleotide position 3685. The valine at codon 1229 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, with isoleucine as the reference amino acid in multiple species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
