NM_007294.4(BRCA1):c.416dup (p.Ser140fs) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.416dupA pathogenic mutation, located in coding exon 5 of the BRCA1 gene, results from a duplication of A at nucleotide position 416, causing a translational frameshift with a predicted alternate stop codon (p.S140Efs*2). This alteration was identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 May;39:593-620). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
