Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_001042492.3(NF1):c.512A>G (p.Asn171Ser), citing Sema4 Curation Guidelines: To the best of our knowledge, the NF1 c.512A>G (p.N171S) variant has not been reported in individuals with NF1-related disease. This variant has been reported in 4/113558 chromosomes in the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 187007). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
