This variant lies in the RAD50 gene (transcript NM_005732.4) at coding-DNA position 3786, where C is replaced by G; at the protein level this means replaces phenylalanine at residue 1262 with leucine — a missense variant. Submitter rationale: The p.F1262L variant (also known as c.3786C>G), located in coding exon 25 of the RAD50 gene, results from a C to G substitution at nucleotide position 3786. The phenylalanine at codon 1262 is replaced by leucine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.005% (greater than 22000 alleles tested) in our clinical cohort. Based on protein sequence alignment, this amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of p.F1262L remains unclear.
