This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 1410, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 470 with aspartic acid — a missense variant. Submitter rationale: The p.E470D variant (also known as c.1410A>C), located in coding exon 13 of the NF1 gene, results from an A to C substitution at nucleotide position 1410. The glutamic acid at codon 470 is replaced by aspartic acid, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6457 samples (12914 alleles) with coverage at this position.<span style="background-color:initial">To date, this alteration has been detected with an allele frequency of approximately 0.002% (greater than 110000 alleles tested) in our clinical cohort.<span style="background-color:initial">This amino acid position is well conserved in available vertebrate species.
